A transient negative shift known as refractory duration prevents the reverse travel of the action potential. In animal cells , two types of these potentials exist, acknowledged, one which is triggered by the voltage-gated calcium channels, the other is caused by calcium channels that are voltage-gated.

In most cells, there is an electrical difference between their surroundings and their interior. The voltage of the cell is measured in millivolts (mV). The voltage of a typical animal cell is -70mV. In the majority of cells few voltage fluctuations are believed to happen, but in a few cells, this is a common feature. Certain cells also exhibit up and down cycles as the voltage goes up and down. The duration of these potentials can vary widely. In brain cells, the up and down action potential cycle is executed in less then a thousandth of a second, whereas in other cells, actions can last for a long time. In the axon hillock, the resting current is around 70 mV and the threshold is 55 mgV. Synaptic connections to the nerve lead to depolarization. This causes the membrane potential to increase or fall. The action potential is generated when enough depolarization is accumulated and the membrane potential reaches an upper limit.

A voltage-gated ion channel is actually a group composed of proteins that remain in the plasma membrane. This produces action potential as it may result in positive feedback loop. Membrane potential is accountable for control of the activity of ion channels. Action potential is generated by the positive feedback cycle that goes on at full-speed. The time and the intensity are determined by the biophysical properties of the ion channel voltage-gated. Ion channels of various types exist that can generate positive feedback mechanisms and create these potentials. Voltage-gated sodium-channels are involved in the generation of higher-frequency potentials that are similar to nerve impulse. In contrast, the slower ones produced in the muscle cells are mediated by the calcium ion channels.The most studied voltage-gated channel are sodium ion channels which are involved in greater speed of nerve impulse conduction.
